SEMで生物試料を観察するために、細胞や生体から摘出した組織などを生きた状態に近い形態に保持すること。
生物は死後の自己融解、さらには生物試料作製過程における脱水や乾燥により変性するため、薬品処理や凍結処理によって、試料を生きた状態に近い形態に保持し、かつSEM観察に耐えられる強度を持たせること。

Fixation is to preserve the morphological structure of biological specimens such as cells and tissues extracted from living organisms, by keeping them close to their living states for scanning electron microscope (SEM) observation.
Since the organisms suffer degeneration due to autolysis after dying and due to dehydration or drying in the specimen preparation process, fixation is carried out by chemical treatment or freezing treatment so that the morphological structure of a biological specimen is preserved close to its living state and the specimen has substantial strength against electron-beam irradiation for SEM observation.
