高分子材料や生物試料など、室温では加工が困難な柔らかい試料から薄膜試料を作製する方法。試料を液体窒素で冷却し、適当な硬さを持たせた後に切削し薄膜試料を得る。切削にはクライオミクロトームを用いる。厚さ数十 nm～数 μmで切り出しが可能である。SEMにおいては、切削後に露出した試料面を観察することもある。

A technique to prepare a thin film from soft materials (macro-molecule materials, biological materials, etc.), which are difficult to cut at room temperature. The specimen is cooled by liquid nitrogen to harden and is formed to a thin section using a cryo microtome. The achievable thickness of the section is several 10 nm to several μm.
It is noted that the exposed faces of the specimen can also be used for SEM observations.
